Wymagania:

We are looking for a highly motivated Postdoctoral Fellow to join the homocysteine pathophysiology research group led by prof. dr hab. Hieronim Jakubowski. The Jakubowski lab looks to uncover the molecular and cellular mechanisms by which dysregulation of one-carbon and homocysteine metabolism accelerates Alzheimer’s disease. We want to learn how homocysteine and related metabolites such as homocysteine thiolactone (HTL) and homocysteine-modified protein (N-Hcy-protein) dysregulate microRNA-dependent epigenetic regulation of mTOR signaling and autophagy, processes that are important for the brain homeostasis and cognition. We use mouse and cellular models of Alzheimer’s disease and a wide array of techniques including analysis of gene expression, RNA and protein chemistry methods, proteomics, chromatin immunoprecipitation (CHIP), immunohistochemistry, confocal fluorescence microcopy, and neurophysiological testing.Applicants should have (1) a PhD degree (either completed or nearly completed), (2) experience in biochemistry, neurobiology, cell biology, or molecular biology, (3) experience in animal models and/or confocal microscopy is welcome, (4) high motivation, initiative, enthusiasm, and readiness to learn new skills, and (5) a record of publication. Opis zadań:

(1) To investigate effects of Blmh deficiency on the development of biochemical and cognitive hallmarks of Alzheimer's disease, using a Blmh-depleted 3xTg-AD mouse model fed with control or hyperhomocysteinemic diet by measuring (1a) cognitive performance and (1b) accumulation of Aβ and pTau deposits in the brain.
(2) To elucidate the mechanisms by which Blmh deficiency can accelerate the development of Alzheimer's disease, by using the Blmh-depleted 3xTg-AD mouse model fed with control or hyperhomocysteinemic diet, as well as by altering Hcy, HTL, and N-Hcy-protein levels in mouse neuroblastoma cells (N2A-APPSwe) to test: (2a) endoplasmic reticulum stress and unfolded protein response, and apoptosis; (2b) neuro-inflammation and immune response; (2c) microRNA and epigenetic regulation of mTOR signaling and autophagy.
