Candidate Profile

- MSc in Physics, Biology, or related fields (completed or near completion)
- Strong interest in interdisciplinary research at the interface of physics and biology
- Experience in fluorescence microscopy, membrane biophysics and/or bacterial culture (advantageous)
- Problem-solving skills and ability to work independently
- Ability to work effectively in a team
- Fluency in spoken and written English
Key responsibilities
- Investigating the role of modified lipid and protein compositions in OMV uptake efficiency by bacterial cells using biochemical approaches.
- Conducting high-throughput and high-resolution microscopy techniques to track OMV interactions with bacterial membranes and cargo release processes.
- Developing and optimizing protocols for the encapsulation of antimicrobial agents within OMV-based carriers, including active loading methods to enhance encapsulation efficiency.
- Performing in vitro assays, including fluorescence assays, and biofilm eradication assays, to quantify OMV delivery efficiency and antimicrobial efficacy.
- Collaborating with external partners for advanced microscopy techniques (STORM, CLEM)

PhD position (m/f/d): Understanding the mechanism of bacterial outer membrane vesicles (OMVs) uptake for targeted delivery
The International Institute of Molecular Mechanisms and Machines (IMol) of the Polish Academy of Sciences invites applications for a 4-year PhD position in the laboratory of Dr. Monika Kluzek.
Project Overview
Gram negative bacteria constantly communicate across one another using outer membrane vesicles (OMVs) and employ them for a plethora of pathophysiological functions within microbial communities. To modulate a response in recipient bacteria, OMVs must be taken up and their biomolecular cargo has to reach the cytosol; how such nanometric vesicles cross the two bacterial membrane layers to finally release their content is still unknown. This project involves using a multidisciplinary approach, combining fluorescence spectroscopy and confocal microscopy, cryoEM and cryoET, and microbiology methods to investigate the mechanism of OMVs uptake by gram-negative bacteria, by focusing on how OMVs membrane biophysical properties regulate the uptake/release process, and how to harness this novel insight to create OMV-based drug carriers for targeted delivery.
