Opis zadań:

Project synopsis:
Recent preclinical data have shown that disulfiram, a potent acetaldehyde dehydrogenase (ALDH) inhibitor used in alcohol use disorder, prevents the development of opioid tolerance, dependence, and withdrawal. The project's overall goal is to identify the molecular mechanisms by which disulfiram and its key end metabolites govern the maintenance of chronic morphine efficacy. The working hypothesis is that modifying specific cysteine residues enhances morphine µ-opioid receptor signaling by activating the Gαi-dependent signaling pathway.
The candidate will be engaged in:
- Implementation of the in vitro morphine-tolerance and withdrawal model
- Detection of the Gi/o signaling pathway proteins and their phosphorylation
- Data analysis, manuscript preparation and submission, presenting results at conferences
